In 643, Monk Jajang (590-658) brought Sakyamuni’s relics from Tang China, which were then enshrined in four temples: Tongdosa Temple in Yangsan, Sangwonsa Temple in Pyeongchang, Bongjeongam Hermitage in Inje, and Beopheungsa Temple in Yeongwol. During the Japanese invasions of 1592-1598, Monk Samyeong (1544-1610) brought some of the relics enshrined at Tongdosa Temple to Jeongamsa Temple. 정암사 적멸보궁은 정선군 정암사에 있는 진신사리*를 봉안한 사찰 건물이다. 이 건물을 지은 시기는 정확히 알 수 없지만, 조선 영조 47년1771에 고쳐 지은 것으로 미루어 보아 18세기 초에 세운 것으로 추정되며 이후에도 여러 차례 보수하였다. 자연석 기단 위에 세워진 앞면 3칸, 옆면 2칸 규모의 건물이다. 겹처마를 드리우고 옆면에도 지붕이 뻗어 있는 팔각지붕을 올려 화려하다.

일반적으로 적멸보궁은 안에 불상을 모기지 않고 근처에 수마노탑을 지어 석가모니의 사리를 봉안한다. 이에 따라 정암사 적멸보궁 안에는 불상이 없고 신중탱화 2점과 동종 1점을 보관하고 있으며 뒷산 중턱에 수마노탑이 있다. 정암사 적멸보궁은 통도사. 법흥사, 상원사. 봉정암의 적멸보궁과 함께 우리나라 5대 적멸보궁 가운데 하나이다.
